RNA is known to associate with chromatin. Various roles for this chromatin-associated RNA have been proposed, including the formation of spatial compartments in the nucleus, recruitment of chromatin modifiers and regulating three-dimensional chromatin organisation. Polycomb Repressive Complex 2 (PRC2) is a crucial repressive chromatin modifier that is known to bind RNA. However, direct roles for RNA in regulating PRC2 catalytic function and chromatin binding are lacking. We have previously shown that an RNA binding surface of PRC2 is required for chromatin modification, but that this activity is actually independent of RNA1. Furthermore, through RNase A treatment of chromatin we uncovered a critical role for RNA in maintaining the solubility and integrity of chromatin2. These findings are consistent with a previously described role for RNA in attenuating electrostatic compaction of chromatin. To further investigate this, we are developing molecular approaches to identify chromatin-associated RNA and its modifications. These approaches are intended to uncover what RNA is localised at specific chromatin compartments, what post-transcriptional modifications this RNA carries and if it has direct roles in regulating chromatin and gene expression.
