Standardised reference materials are essential for ensuring consistent evaluation of mRNA therapeutics across sequence design, manufacturing, quality control, and functional assessment. We developed a universal mRNA reference standard that supports benchmarking throughout the complete mRNA development workflow. A foundational reference sequence was generated to enable comparison of mRNA design algorithms, while additional reference materials were applied to establish and validate analytical assays for critical quality attributes, including transcript integrity, double-stranded RNA content, 5′ capping efficiency, and poly(A) tail length. Standardised cellular assays were also implemented to assess how design and manufacturing parameters influence mRNA expression and stability. Collectively, these reference standards provide a comprehensive framework for evaluating process improvements and technological innovations across the mRNA lifecycle, enabling consistent comparison of their effects on final product quality and performance.
